In the article by MOIRA M. COCKELL, SEVERINE PERROD and SUSAN M. GASSER (Genetics 154: 1069–1083) entitled "Analysis of Sir2p Domains Required for rDNA and Telomeric Silencing in Saccharomyces cerevisiae," the plasmids used in this study carrying the full-length or core SIR2 gene labeled as sir2-394L have point mutations that result in the substitution of amino acid residues Arg403 and Arg404 by glycines, in addition to the reported replacement of the conserved proline residue at amino acid 394 by leucine. The Pro394 to Leu mutation alone eliminates Sir2p function and confers a dominant negative phenotype for telomeric silencing similar to that reported. A detailed analysis of differences contributed by these two additional mutations will be presented elsewhere.
